Preventive approach against drug-induced pulmonary fibrosis through the suppression of epithelial-mesenchymal transition

چکیده
A number of drugs induce pulmonary injury and subsequently lead to serious lung diseases such as fibrosis the adverse drug reactions. However, an effective preventive approach against drug-induced has not been established due poor understanding common targets in a variety showing toxicity. Epithelial-mesenchymal transition (EMT), cellular phenotypic change epithelial mesenchymal state, contributes development through conversion damaged alveolar epithelium into myofibroblasts. As several with toxicity have reported EMT, EMT serves bridge between fibrosis. Accumulated evidence supports potential target Additionally, since there are mechanistic differences main pharmacological effect induced by drug, prevention based on suppression would be possible contribute continuous clinical treatment avoid EMT-mediated Furthermore, targeting seems adequate for exerting cells occurs prior pathophysiological state whole bleomycin-induced rat model. This viewpoint deals benefits perspectives approaches druginduced which rarely addressed.

Nintedanib reduces ventilation‐augmented bleomycin‐induced epithelial–mesenchymal transition and lung fibrosis through suppression of the Src pathway
Mechanical ventilation (MV) used in patients with acute respiratory distress syndrome (ARDS) can increase lung inflammation and pulmonary fibrogenesis. Src is crucial in mediating the transforming growth factor (TGF)-β1-induced epithelial-mesenchymal transition (EMT) during the fibroproliferative phase of ARDS. Nintedanib, a multitargeted tyrosine kinase inhibitor that directly blocks Src, has ...
متن کاملProtective Effects of Hydrogen-Rich Saline Against Lipopolysaccharide-Induced Alveolar Epithelial-to-Mesenchymal Transition and Pulmonary Fibrosis
BACKGROUND Fibrotic change is one of the important reasons for the poor prognosis of patients with acute respiratory distress syndrome (ARDS). The present study investigated the effects of hydrogen-rich saline, a selective hydroxyl radical scavenger, on lipopolysaccharide (LPS)-induced pulmonary fibrosis. MATERIAL AND METHODS Male ICR mice were divided randomly into 5 groups: Control, LPS-treat...
متن کاملSaffron Protection against Bleomycin-Induced Pulmonary Fibrosis in Rats
Background: Bleomycin-induced lung fibrosis has been accepted as an animal model for fibrosis in rats. The aim of this study was to evaluate the effects of saffron aqueous extract on this disorder paving the way for more investigation in treating idiopathic pulmonary fibrosis in human. Methods: Male Wistar rats (250–300 gr) were instilled a single dose of bleomycin (5 mg/kg) via in...
متن کاملLysyl oxidase promotes epithelial-to-mesenchymal transition during paraquat-induced pulmonary fibrosis.
Lysyl oxidase (LOX) is a copper-dependent amine oxidase that plays a critical role in pulmonary fibrosis. Our previous study demonstrated that epithelial-to-mesenchymal transition (EMT) was strongly associated with paraquat (PQ) induced pulmonary fibrosis. This present study was aimed to evaluate the potential involvement of LOX on EMT in the process of pulmonary fibrosis induced by PQ. We esta...
متن کاملEmodin ameliorates bleomycin-induced pulmonary fibrosis in rats by suppressing epithelial-mesenchymal transition and fibroblast activation
Aberrant activation of TGF-β1 is frequently encountered and promotes epithelial-mesenchymal transition (EMT) and fibroblast activation in pulmonary fibrosis. The present study investigated whether emodin mediates its effect via suppressing TGF-β1-induced EMT and fibroblast activation in bleomycin (BLM)-induced pulmonary fibrosis in rats. Here, we found that emodin induced apoptosis and inhibite...
